Overview
CJC-1295 (DAC) is a synthetic 30-residue analog of growth hormone-releasing hormone (GHRH 1-29) incorporating four amino acid substitutions and a C-terminal lysine bearing a maleimidopropionyl group. This Drug Affinity Complex (DAC) moiety forms a covalent bond with cysteine-34 of circulating albumin after administration, extending the analog's half-life from minutes to several days. Developed by ConjuChem researchers in the early 2000s, it is used as a reference long-acting GHRH receptor agonist in somatotropic axis research.
Mechanism of action
Activates the GHRH receptor, a class B G-protein-coupled receptor expressed on somatotroph cells of the anterior pituitary. Receptor activation couples to Gs, raising intracellular cAMP and activating protein kinase A, which increases growth hormone gene transcription, somatotroph proliferation, and calcium-dependent hormone release. The substitutions at positions 2, 8, 15, and 27 confer resistance to DPP-4 cleavage at the N-terminus and reduce oxidation and rearrangement. Albumin conjugation via the DAC linker limits renal clearance and protects the sequence from proteolysis, producing sustained receptor exposure while endogenous somatostatin tone continues to shape pulsatile release.
Key research findings
- 01
Rat studies identified CJC-1295 as the lead albumin-reactive GRF conjugate, demonstrating sustained pituitary GRF receptor activation compared with the unconjugated analog.
- 02
Clinical studies in healthy adults characterized dose-dependent elevations in mean growth hormone and IGF-1 concentrations persisting for days after a single administration, with an estimated half-life of roughly six to eight days.
- 03
Frequent-sampling studies demonstrated that pulsatile growth hormone secretion persists during continuous stimulation, with increased trough levels rather than abolished pulsatility.
- 04
Studies in GHRH knockout mice characterized normalization of linear growth and body composition endpoints under repeated exposure.
