§ Research
CJC-1295/Ipamorelin is a combination of two growth hormone secretagogues acting through distinct receptors. CJC-1295 is a tetrasubstituted analog of growth hormone-releasing hormone (GHRH 1-29), available with or without an albumin-binding Drug Affinity Complex. Ipamorelin is a synthetic five-residue ghrelin receptor agonist noted for its selectivity for growth hormone release. The pairing is used in research examining the interaction between GHRH receptor and ghrelin receptor signaling in the somatotropic axis.
The CJC-1295 component activates the GHRH receptor, a Gs-coupled class B G-protein-coupled receptor on anterior pituitary somatotrophs, increasing cAMP, protein kinase A activity, growth hormone gene transcription, and hormone release. The ipamorelin component activates the growth hormone secretagogue receptor type 1a (GHS-R1a), a Gq-coupled receptor in the pituitary and hypothalamus, mobilizing intracellular calcium via phospholipase C and functionally opposing somatostatin tone. Because the two receptors use separate second-messenger systems, co-activation is reported in the component literature to produce greater growth hormone release than either pathway alone.
Clinical studies of CJC-1295 in healthy adults characterized sustained, dose-dependent elevations in growth hormone and IGF-1 concentrations following single and repeated exposure.
Preclinical swine and rat studies characterized ipamorelin as a selective secretagogue that releases growth hormone without significant elevation of ACTH or cortisol, unlike earlier GHRP-class compounds.
Classic pituitary studies established that GHRH receptor agonists and ghrelin receptor agonists act synergistically on growth hormone release, providing the pharmacological rationale studied for such pairings.
The combination itself has not been characterized as a single entity in peer-reviewed studies; available data derive from published literature on the individual components.
